PluriKit™: End-to-End Solution for Consistent iPSC Growth

Achieving reproducible iPSC growth has long been a challenge, especially with the availability of various reagents and methods creating endless combinations. To address this, Pluristyx has developed PluriKit, an integrated solution specifically designed to support the reproducible culture of PluriBank™ RUO FailSafe® lines.

We evaluated and picked the best performing reagents to produce a robust and cost-effective growth paradigm. This ensures proper culture and cryopreservation of iPSCs while maintaining viability, pluripotency and genomic integrity of the cells.  We include in the bundle an iCS digital™ assay to be run on the first expanded bank of cells.

Reagents Selected for Optimal iPSC Growth

• Laminin-521: A reliable cell culture substrate for consistent adherence and growth.
• HiDef-B8 Medium: A modified version of Essential 8
• PBS-EDTA: A gentle effective cell dissociation reagent for clump passaging.
• CEPT Cocktail: A blend of pro-survival molecules as an alternative to ROCKi.
• PluriFreeze 10: A cryopreservation reagent developed by Pluristyx, specifically tailored for pluripotent stem cells.

Proven Performance

The PluriKit™ system is designed specifically for PluriBank™ RUO FailSafe® lines to achieve:

• ≥ 8-fold growth over 4 days.
• 18–24-hour doubling time.
• ≥ 70% viability at harvest, with normal karyotype retention.
• Supports both daily and skip-day feeding paradigms.
• Includes a genetic testing package for the first cell bank made from Stem Genomics.
